   Proposal Description:
   Obesity is a risk factor for poor neurocognitive outcomes, including              Alzheimer's disease (AD). Recent work demonstrates that obesity is associated     with AD-related neuropathological changes however, the sample sizes have been     relatively small and the mechanisms for this relationship remain unclear.         Obesity is associated with cerebrovascular disease and micro- and microvascular                                                                                   disease have been suggested to play a key role in the pathogenesis of AD,         suggesting that cerebrovascular alterations may be critical in obesity-related    AD-risk.                                                                                                                                                            The purpose of the proposed study is to first exploit the large NACC dataset to                                                                                   examine the association between body mass index (BMI) and AD-related              neuropathological changes in patients with AD. We will then investigate whether                                                                                  cerebrovascular disease, as operationalized via the Framingham Stroke Risk        Profile and cerebrovascular neuropathology, mediates the association between      BMI and AD neuropathology.                                                                                                                                          The proposed study will test the hypothesis that higher BMI will predict          greater AD-related neuropathological changes that will be mediated by             cerebrovascular disease. The proposed study will be the first to examine BMI      and AD neuropathology in a large population-based sample and to also determine    the mechanistic pathways into the relationship between BMI and AD                 neuropathology.
